Transgenic zebrafish indicating environmental estrogen pollution was successfully constructed in Fudan University

A transgenic zebrafish, which indicates directly environmental estrogen pollution, was constructed successfully in Fudan University , and it was validated by experts. Professor Houyan Song, the academic chair of the Key Laboratory of Molecular Medicine, Ministry of Education, Fudan university, and special appointed Professor Tao Zhong have been in charge the program. It is funded by Shanghai natural science foundation.

In recent years, there is a considerable concern regarding environmental estrogens (EEs). These chemicals possess the capacity to modulate endocrine system and affect reproductive health in wildlife, even in humans. EEs are widespread in water, food and soil. The effects of EEs have been observed in all classes of vertebrates. It was estimated that even very low concentration of EEs impair male reproductive health. In the past 50 years, it was reported that the total sperm numbers and sperm volumes have reduced by 40% and 20% in men, respectively. EEs lead to poor fecundity, increase rate in congenital abnormality and may associated with the elevating rate in testicle tumor and prostate cancer.

An automatic scale up zebrafish facilities has been established. The zebrafishs with clear genetic background were imported 3 years ago from USA . Based on the science and technology platform of the zebrafish, a Vtg promoter which is sensitive to estrogens was cloned and identified. An EGFP expression plasmid driven by this promoter was then constructed and micro-injected into zebrafish fertilized eggs. After a large scale screening, the F1 generation transgenic zebrafish sensitized to EEs was identified eventually. The transgenic zebrafish is able to demonstrate aquatic estrogen pollutions directly and sensitively. It was reported that even there is a trace of estrogens in the water, transgenic zebrafish liver exhibits green fluorescence.

2 national invention patents, and 8 papers related to the transgenic zebrafish were applied or published in international journals. A minitype automatic zebrafish facility invented by the Key Lab was also applied for a nation patent. Experts said it is quite a convenience using the transgenic zebrafish to test aquatic estrogen pollution, and it has lots of theoretical and applied values in environmental protection, reproductive and pathological researches.
